青い瞳の浪人・眠狂四郎（長谷川博己）は、老中水野忠邦の側近・武部仙十郎（宅麻伸）の依頼で、忠邦の命を狙う間者を斬り捨てる。間者の正体を探るうち狂四郎は間者の妹で切支丹の茅場静香（黒島結菜）と運命的に出会うが、隠れ切支丹の頭目・備前屋（神保悟志）は信徒たちの存在を知った狂四郎の暗殺を仲間に告げる。一方大目付・松平主水正（坂東彌十郎）は、腹心・戸田隼人（高橋光臣）に狂四郎の命を狙うよう命じていた。
